Caskey is a principal of the Washington Advisory Group. He was on the American Cancer Society's Advisory Committee on Nucleic Acids and Protein Synthesis in 1974. Other top ACS officials included Lasker Foundation director and WAG principal Purnell W. Choppin; Council for Tobacco Research members Wolfgang Joklik, Peter K. Vogt, and Roswell K. Boutwell; Frank Rauscher Jr. of the NCI; and R. Lee Clark of MD Anderson.

Caskey was on the Board of Trustees of the Cold Spring Harbor Laboratory (founded by James D. Watson), in 1996 and 1997. CSHL published the 1980 Banbury Report 3, a red herring created by ACS and NCI insiders including Ernst L. Wynder of the American Health Foundation and Gio Bata Gori of the NCI. It was also the location of the founding of the Human Genome Organization (HUGO).

Caskey got his MD at Duke University in 1963. He was president of the Human Genome Organization (HUGO) from 1993 to 1996. From 1994 to 2000 he was Senior Vice President of Research at Merck Research Laboratories, and Trustee and President of the Merck Genome Research Institute from 1999-2000. He is on the Intramural Human Genome Projects Special Review Committee at the National Institutes of Health; is on the editorial boards of the Journal of the American Medical Association and Science; and is a member of the Institute of Medicine. He is or has been on the boards of numerous corporations, including Athersys; Dynagene; Genometrix; Lexicon; Luminex; Odyssey; Vical; and Xeotron.

Caskey was chairman of the board of Lexicon Genetics from 2000 until 2005.

Caskey joined the board of directors of Luminex Corporation in 2001. Former CDC Director William L. Roper has been a director of Luminex since 2000.

Caskey is on the Review Panel of Genome Canada.
